Description of the department and the scientific environment
Department of Clinical Immunology is the regional department for Clinical Immunology and Transfusion Medicine. The department carries out immunological diagnostics, including inborn immunodeficiencies and autoimmune diseases as well as blood bank management and production of blood products. Research is an integrated part of the department and we have a dedicated group of MSc’s, medical doctors and bioengineers involved in our research group. The research area covers autoimmune diseases, immunodeficiencies, genetic association studies, extracellular vesicles and cancer immunotherapy. The department has national reference laboratory status for investigation of immune neutropenia’s.
The department has about 105 employees, including two associate professors, eight medical doctors and six researchers. The department is equipped with modern instruments for molecular and cellular analysis. The department will be located in the New Aalborg University Hospital from 2022.
The positions and project descriptions
The research is founded in our existing research in primary immune neutropenia in children. The PhD. fellow will be in close collaboration with paediatric departments. On daily basis, the student will be associated to our molecular biology laboratory. For the right person, there will be possibilities for a post doc position following the PhD project.
The PhD. project
A study of immune regulatory gene polymorphisms and FCGR3B copy number variation in relation to pathognomonic autoantibodies in primary autoimmune neutropenia of infancy.
Background
Primary autoimmune neutropenia of infancy and childhood (AIN) is the most frequent cause of severe neutropenia in children and can lead to severe bacterial infections. The disease is caused by autoantibodies directed against membrane proteins on neutrophil granulocytes. The antibodies can be directed against HNA (Human Neutrophil Antigen) antigens, most frequently epitopes on the fc gamma receptor 3 (bFcγR3b), a receptor involved in immunoregulation, and encoded by the FCGR3B gene. The etiology of AIN has yet to be elucidated and the genetic background for the disease remains unknown. The goal of this study is to investigate inborn genetic variation in children with AIN.
Main objectives of the study
- Establish an assay and determine association between FCGR3B copy number variation and AIN
- Investigate the association between 50 single nucleotide polymorphism ( SNPs) in immune regulatory genes and AIN
- To compare a novel and rapid method for detection of antineutrophil antibodies: LabScreen MULTI (One Lambda Inc.) with a standard flow cytometric method and investigate the asociation to genetic risk factors.
