We are seeking a highly motivated researcher who will focus on characterizing how key metabolites such as succinate and lactate function as extracellular signals of metabolic stress or fuel availability through recognition by and signaling through specific GPCR sensors. An important part of the research will be to translate cutting edge basic research in this field into innovation. The position is a 2 year position to start January 1, 2022 or after agreement.

Background
The Novo Nordisk Foundation Center for Basic Metabolic Research’s vision is to strengthen interdisciplinary research that transforms our basic understanding of the mechanisms that underlie metabolic health and disease, and to accelerate this knowledge towards new prevention and treatment strategies. We were established in 2010, and in 2017, we moved into new laboratories, offices and meeting rooms in the Maersk Tower www.maersktower.ku.dk.

Our group and research
The aim of the Schwartz Group is to understand the structural and functional basis for how signaling metabolites and their specific GPCR sensors control endocrine, metabolic and immune functions as the basis for developing novel means to prevent and treat obesity, diabetes and cancer and to translate this research into innovation projects. Project description
Over the last five to eight years, we have established that key metabolites from our intracellular intermediary metabolism function not only as fuel add metabolic building blocks but also act as extracellular signals of fuel availability and metabolic stress.

The specific project mainly deals with:

  1. characterizing how succinate is generated and accumulates in response to metabolic and hypoxic stress and how it binds to and activates different signal transduction pathways through its GPCR sensor, GPR91/SUCNR1 and
  2. characterize how lactate as an extracellular endpoint of glycolysis bind to and activates GPR81/HCAR1. These two integrated projects both covers auto and paracrine mechanism in metabolic tissues, immune cells and cancer and the work spans from basic molecular pharmacology - including participation in the development of novel pharmacological tool compounds – over in vitro 2D and 3D cellular models to in vivo transgenic and syngeneic murine models. Importantly, at this point the project has matured to also function as the basis for an innovation project aiming at forming a start-up biotech company.

Your job
You will function as the right hand of the Group Leader, Thue W. Schwartz both in respect of running the day to day activities in the basic research project and in the innovation project as these are still closely integrated. You will coordinate the day to day work of two lab technicians and one PhD student, and act as project coordinator for the very interdisciplinary projects including close interactions with a neighbouring computational/medicinal chemistry group, a cancer group as well as several other collaborators including CRO’s.

Profile
A highly motivated and enthusiastic researcher with a PhD within molecular and cellular biology, who based on the fact that this is an ongoing major project should have the following competencies and qualifications:

Essential criteria:

  • Deep knowledge of GPCR related molecular pharmacology including hands-on experience with advanced signal transduction assays (minimum including Ca2+, IP3, cAMP, G protein recruitment (BRET based assays), luciferase assays).
  • Experience with structure-based discovery and characterization of mode of action of pharmacological tool compounds. Hands on experience with screening small molecule libraries.
  • Extensive experience with isolation, differentiation and characterization of both primary human and murine immune cells
  • Solid experience with molecular biology (cloning, mutagenesis, qPCR) and cell biology techniques (including experience with culture and handling of multiple cell lines as well as primary cells, and characterizing them with IncuCyte®, Seahorse® and xCelligence® equipment for real time analysis).
  • FELASA laboratory animal science certificate and hands-on experience in applying for licenses for animal experiments, handling transgenic mouse model breeding etc. and experience in planning and running various syngeneic cancer models.
  • Basic training in the underlying mechanisms and key concepts within the innovation process behind starting up a biotech company.
  • Experience in day to day supervising master and PhD students as well as lab technicians. Proven scientific track record.
  • Excellent skills and training in presenting data at meeting

Relationships:

  • The holder of the position will report to Professor Thue W. Schwartz.
  • It is key that the holder of the position excel in interacting with staff at all levels, both internally and externally, regarding relevant research topics.

Place of employment
The place of employment is at The Novo Nordisk Foundation Center for Basic Metabolic Research, Maersk Tower, Panum Institute, University of Copenhagen. We offer creative and stimulating working conditions in a dynamic and international research environment.

Terms of employment
The employment as postdoc is a 37 working hours (full time employment) and fixed-term position for 2 years. Starting date is January 1, 2022.

Salary, pension and terms of employment will be in accordance with the agreement between the Ministry of Finance and AC (Danish Confederation of Professional Associations). Depending on qualifications, a supplement may be negotiated.

Non-Danish and Danish applicants may be eligible for tax reductions, if they hold a PhD degree and have not been tax liable in Denmark the last 10 years.

The position is covered by the “Memorandum on Job Structure for Academic Staff at the Universities” of December 11, 2019.
