This Postdoc position is associated with the ERC Consolidator project “The biofilm matrix and its functional role in the ecology of bacterial communities – BioMatrix. The overall aim of the project is to examine the production and composition of matrix components in multispecies biofilms and their role in shaping community function, activity and protection. A four-species soil community interacting synergistically in biofilm formation will be used as model. Experimental designs will span various levels of community and environmental complexity to facilitate both a mechanistic and an integrative understanding of the impact of the biofilm matrix for community ecology.

The main task of the Postdoc fellow will be to genetically modify the model strains based on traditional cloning protocols and novel, alternative approaches including alternative tools (e.g. the CRISPR-Cas system). The aim is to analyze the spatiotemporal expression of specific matrix components and their impact for biofilm functionality. Specific tasks include 1) tagging of strains with fluorescent markers to allow analysis of spatial organization using confocal laser scanning microscopy, 2) generation of knock out-mutants with specific deletions of genes/operons encoding matrix components and 3) construction of biosensors reporting the expression of specific matrix components. Engineered strains will then be introduced to various in vitro model systems followed by structural and functional characterization of the community.

The Postdoc position is for 2 years and the project will be carried out at Section of Microbiology, Department of Biology, University of Copenhagen where you will engage in teamwork within the group and with national and international collaborators.

The position is open from 1 September 2021.

Qualifications

We are looking for a highly motivated and ambitious researcher with strong experience in biofilm microbiology, genetic engineering and bacterial interactions. Applicants should have the following qualifications:

  • PhD in Microbiology, Bioengineering, Molecular Biology or equivalent
  • Hands on and documented experience with cultivation and analysis of bacteria
  • Hands on and documented experience with genetic engineering of bacteria and fluorescence-based reporter systems.
  • Previous experience with use of in vitro biofilm models and confocal microscopy analysis of bacterial communities
  • Previous experience with genetic tools and engineering in Gram positives and/or alternative genetic engineering tools (e.g. the CRISPR-Cas system) will be seen as an advantage
  • Fluency in English

Main tasks of the appointed applicant

The postdoc’s duties will include research within microbiology as well as teaching. Tasks include:

  • Genetic engineering of bacterial model strains
  • Visual analysis of spatial community structure using confocal microscopy
  • Structural and functional characterization of the biofilms using e.g. stains binding specific matrix components, biomass and viability assays etc.

The post may also include performance of other duties.

Terms of employment

The position is covered by the Memorandum on Job Structure for Academic Staff.

Terms of appointment and payment accord to the agreement between the Ministry of Finance and The Danish Confederation of Professional Associations on Academics in the State.

The starting salary is currently up to DKK 440,935 including annual supplement (+ pension up to DKK 75,399). Negotiation for salary supplement is possible.
