Frederiksberg C

We are seeking a highly motivated postdoctoral fellow for a project centered on the development of a yeast single-cell mucus layer model, using microfluidics and yeast biosensors designed for human sensory receptors.

The roles will mainly be dedicated to two lines of work:

  1. Implementation of standard molecular biology methods for engineering yeast cells at the Department of Food Science. Yeast expression of human sensory receptors in a MAPK biosensor chassis using a fluorescence readout, activity assays, and fluorescence microscopy to measure activity and surface exposure.
  2. Develop and explore the feasibility of a two-phase microfluidic model system for yeast biosensors.

In addition to the core research lines mentioned above, the postdoctoral fellow will participate in and present research during the regular research group meetings and the bi-weekly flavor chemistry group meetings.

The research

Sensing of aroma compounds is a complex process that includes several specialized cell types for structure, electrochemical potential, and maintenance of the homeostasis in the olfactory mucosa. This research area is focused on understanding how the physiological conditions of the nasal cavity and mutations in the olfactory receptors affect their activity. The yeast biosensor for human GPCRs will potentially allow massive parallel screening of mutations in human populations, and the microfluidic system will be the first model system for native physiological conditions. This research is branching off our long-lasting track record of working with sensory science across age differences, cultures, cultures, and disease, trying to understand the differences we observe in sensory experiments but cannot explain. Adapting the conditions to disease and aging will help us understand the mucosal microenvironment's impact. You will be pioneering these experiments with the aid of experts in the field. Our research facilities include modern sensory and flavor chemistry laboratories, which are part of the Future Consumer Lab (https://futureconsumerlab.ku.dk/). The Department of Food Science has facilities for cell manipulation and equipment at the Microbiology department and access to the Center for Advanced Bioimaging (https://cab.ku.dk/) at our campus for fluorescence microscopy. The microfluidic development will take place in the Microscale Analytical Systems group (https://pharmacy.ku.dk/research/microscale-analytical-systems/) at the Department of Pharmacy.

Profile

The successful candidate will take initiative and responsibility, work independently, and thrive in this highly collaborative framework. We are looking for a highly motivated and enthusiastic scientist with the following competencies and experience:

Essential experience and skills:

  • You must have extensive experience with yeast manipulation and molecular biology, including cell transformation, yeast culturing methods, growth measurements, and plasmid construction.
  • You have a PhD in Molecular Biology, Biotechnology, Biochemistry, Bioengineering, or related sciences
  • You embrace technological challenges
  • Excellent English skills, written and spoken.
  • Good dissemination skills for internal talks and external community via conferences and publications.

Practical experience with the following techniques is favored but not a necessity

  • Microfluidics
  • Fluorescence microscopy
  • Expression of fluorophores in yeast.
  • Human physiology and biochemistry.

Place of employment

You will be employed at the Department of Food Science, University of Copenhagen, under the supervision of Tenure-track assistant professor Sylvester Holt, and in the research group of Microscale Analytical Systems at the Department of Pharmacy, in the laboratory of Professor Jörg Kutter.

Terms of employment

The average weekly working hours are 37 hours per week.

The Velux Foundations are funding the project, with a current duration limit of 2 years. We encourage the postdoctoral fellow to apply for future funding. The desired starting date is February 1, 2024.

Salary, pension and other conditions of employment are set by the Agreement between the Ministry of Taxation and AC (Danish Confederation of Professional Associations) or other relevant organization. Currently, the monthly salary starts at 36,400 DKK/approx. 4,880 EUR (October 2023 level). Depending on qualifications, a supplement may be negotiated. The employer will pay an additional 17.1 % to your pension fund.

Foreign and Danish applicants may be eligible for tax reductions if they hold a PhD degree and have not lived in Denmark for the last 10 years.
